That first exclusion is the one that catches people. The bus stops outside wineries; the bus ticket does not buy you anything inside them. One reviewer describes the day well: “Tasted wine at the Batch Winery. Olive oil and honey at Allpress. Both excellent.” (Elaine). She tasted, she enjoyed it, and she paid for it separately. Budget for that or you will spend the day looking at gates.
NZ$99 in the combo, NZ$59.50 on its own
The bus pass by itself is NZ$59.50 for an adult. Bundled with the return ferry it is NZ$99, which prices the crossing at NZ$39.50 — less than the NZ$46.50 off-peak return and a long way under the NZ$62 on-peak one. That is the whole argument for the combo, and it is a good one if you were going to ride the bus anyway. A reviewer puts it plainly: “Paid less than purchasing trips separately. All staff were friendly, helpful and welcoming.” (Loretta Kathleen). Child is NZ$51 and a family of two adults and two children is NZ$209. The catch is timing. Another reviewer on this exact product writes: “I HIGHLY recommend getting an early ferry so you have time to get off the hop on/hop off tour bus and explore” (Kiana). She is right, and an early sailing is an on-peak sailing. The combo and the off-peak saving pull in opposite directions; you cannot have both.

The e-bike looks NZ$20 dearer than the combo and is really NZ$66.50 to NZ$82 dearer, because it starts at Matiatia and leaves you to buy the crossing. It is also the one with genuine conditions attached: ages 12 and up, 145cm minimum height, and you have to be able to ride. Only 35 people have reviewed it, and the four-star reviews are the useful ones — “It would have been nice to have information about the length of the routes with the corresponding battery before booking” (Ute). Another rider is blunter about who it suits: “I definitely recommend this for anyone who regularly cycles 50+ kilometers.” (Nienke). The island is 19.3 km long and hilly, topping out around 231 metres, so take her seriously.
The third option is the one nobody sells you. There are public buses on Waiheke run under Auckland Transport. They are not free, and I am not going to quote you a fare I could not verify from a primary source — check Auckland Transport directly. Taxis operate on the island too. What I would not do is plan a day around getting a rideshare: coverage is unreliable to absent and visitors report not being able to get one.
If the point of your day is wine rather than transport, the bus is the wrong purchase and a guided day is the right one. The full ladder of tickets and tours puts both in price order.

Does the hop-on hop-off bus include wine tasting?

Is the hop-on hop-off bus worth it?
If it is your first visit, you have no plan and you want to see several bays without organising anything, yes. If you are confident on a bike, comfortable working out a public bus, or you only want one village and one beach, you are paying for stops you will not use. The island is 19.3 km long and hilly, so walking between stops is not the fallback people assume — see whether you can walk around Waiheke.

The Auckland Ferry Terminal at 99 Quay Street, Downtown Auckland, arriving at Matiatia on Waiheke in about 40 minutes. The bus meets the ferry at Matiatia. Sailing times are seasonal, so check the operator’s live timetable rather than any list on a guide site, including mine.
